All of a sudden we're finding that customers can't login to our store. They are not being recognised by the system even though their details are in admin. I tried doing a password resend, but the customer email address isn't recognised either.

Something has changed the Matrix.
Either a change was made by your Hoster or to your files. Find out if Server software was recently upgraded and start looking at error logs. Your Hosting account should have an error log you can view and there is a Debug Error Logging module you can download from the Add Ons section.
I had the same issue some time before.
Please check your cache settings (clear it), when it is full or when your cache table in the database is corrupted, the login session / cookie cannot be set if you use database as your session catcher. .
I my case, just before I jumped out of the window, it worked. after 1 day later i tried it again and then everything worked fine . I guess my hoster was updating / moving something without notifying anyone.

I'm having the same issue. I just upgraded to the latest version of zencart.
Any resolution on this? How do I clear my cache in Zencart?
