Many hosts prevent FTP programs from setting files lower than 644, but that still leaves the file writable.
So, you need to use the host's control panel file manager to set the permissions lower.
That's all described in this FAQ article: https://www.zen-cart.com/tutorials/index.php?article=9

I am having an issue with a site that is switching the file permission of /includes/configure.php to CHMOD 600

Even had my hosting company on to it and they are at a loss as to why we switch to either 444 or 644 and within minutes it's back to 600.

Unless you've added some code to do it, Zen Cart DOES NOT change permissions on files to 600.
Best to work with your hosting company to figure out what's causing the change, and/or inspect your Zen Cart files for alterations you've made/added since the original files were obtained.
