Changes to layout boxes are not updating. Why?

I'm using the latest version with no addons and no customizations. The site is hosting on HostGator.

Why are the changes not being reflected?

Please elaborate on what changes, and how you are attempting to do them - in detail

I click the green e icon to edit any of the layout boxes and then click the grey edit button. When I toggle the on/off LEFT/RIGHT COLUMN
Status and choose "Update" nothing is happening. Incidentally, there isn't a confirmation or error message.

Have you installed the security patches? Admin changes that don't 'take' is one possible symptom of security patch files being uploaded to the cart folders rather than the admin folders, where they belong.

If that isn't it, try re-uploading the admin folder and all its contents.
