The only file in the custom directory is header.php - it is the file that I edited for the new logo and template. Should I delete it?

I should add that I copied the header.php file originally from the includes/languages/english directory so I could put the name of the logo file in it.
I deleted the custom folder and still got the same error. I think I will undo all the template mods and start over.
I deleted all the template mods and now I can use the template selector and the 500 error is gone - probably one of the files was corrupted or had bad code. I wll start this mod again be a bit more careful. Thanks for the help (and idea about the template selector).
I noticed you said the image was a png..... the header.php file has to be changed to reflect this association change. Default has it set as logo.jpg so you would need to edit that line to reflect you are using a png IE.. logo.png

I deleted all the entire folder. I must have missed a step which caused my problems so I will go back to the beginning. Thanks to everyone who responded.
If you view the image properties on your site, example on windows: right click > properties you can see the image path, replace the image by following that path.
