Are you using Image Handler? If so, that breaks a whole load of other contribs, including lightbox, as IH hasn't been updated in a LONG LONG time.

If you are, then backup, remove IH from the database, delete all remnants of it, replace the files with stock zen files and lightbox should start working. It worked for me...
