I get this all the time since i set the permissions on my define files to read only. So I just ignore those errors when using the devlopers toolkit and I just change the permissions on those files to 777 when editing and back to 644 after I'm done.

if you change the files chmod to 777 when editing or using the developers tool kit the error should go away, but I wouldn't leave there I would always set the define files to 644 or lower if you can after your done editing them. The directory setting is fine.

Generally speaking, 744 is as secure as 644 with respect those files.
