At a technical level, that message literally means that when PHP checked to see if the path entered could be found on the server ... that it didn't find it, so a "failed" result came back.
Does it make any difference if you add another / to the end of it ?.

At the start i get these also

What do these mean??
Also when i put the / at the end of the
e:/domains/p/puffyourstuff.co.uk/ still doesnt work, I really need help
Im using Streamline.net for my hosting.

What PHP version is running on the server?
Do you know what version of IIS and Windows Server are running on it?
Is there a particular reason why you're choosing windows hosting instead of linux?.
