I believe this is a very unusual error; whenever an attempt is made to change define_main_page through admin > tools > Define Page editor, the following error appears, regardless of whether I'm using an editor or plain text:I know WHAT is causing the problem, but I don't know why, because it hasn't caused a problem for the past 18months!Not Acceptable

406 errors are typically a result of mod_security rules blocking some or all of the submitted content.
Your server's errorlog should tell you more about which specific rule is involved..

Interesting - I've just tried to run an sql query through admin > tools > install sql patches and got the same warning - on a completely different server on a different host.

Same issue. Probably a slightly different mod_security "rule", but still likely a mod_security thing.
